Protocol for formulating ABATE 4E for larval bioassay

ABATE 4E contains 44.6% temephos. Take 2.24 ml of "off-the-shelf" ABATE 4E and pipet it into a 100 ml volumetric flask. Fill to 100 ml with absolute ETOH. This is STOCK solution 1.

Take 1 ml of STOCK 1 and put it into a 100 ml vol. flask. Fill to 100 ml with absolute ETOH. This is STOCK solution 2.

Take 1 ml of STOCK 2 and put it into a 100 ml vol. flask. Fill to 100 ml with water. This is the TEST solution. The TEST solution is 1 ppm. Be sure to use water as the solvent in the preparation of the TEST solution

For the bioassay, we will try 1/2 ml, 1 ml, 2 ml, 3 ml, 4 ml, etc. in 250 ml final volume plastic cups. We will test 10-15 3rd instar Culex pipiens larvae in each test cup. Final volume is 250 ml.

Pipette ½ ml, 1 ml, 2 ml, and 4 ml of water into each of 4 CONTROLs.

In 24 hours, score the percent mortality and analyze by probit analysis. We use POLO PC.
